Analysis of Covid-19 infection and inflammatory response on a fully differentiated human bronchial epithelium model.

C’est suite à plusieurs semaines de travail et de concertation que 2 équipes de l’unité, Andevir dirigée par M-L. Andreola (porteur du projet) et SpacVir dirigée par H. Wodrich et M-E. Lafon avec la collaboration d’une troisième équipe, T. Trian (CRCTB, INSERM U 1045) ont obtenu un financement de 18 mois pour la recherche contre le SARS-CoV-2. Dans ce consortium sont également impliqués le service des maladies infectieuses et tropicales du Pr. D. Neau (Pr D. Malvy) ainsi que le service de médecine intensive – réanimation du CHU (Pr A. Boyer).

In this project, we aim to assess the SARS-CoV-2 infection in a primary bronchial epithelium model. Using this highly relevant and physiological model we will employ imaging, RNA analysis and cytokine profiling to determine i) which cell type is (are) the primary target of SARS-CoV-2 and ii) what is the contribution of the bronchial epithelium to the immune response triggered by SARS-CoV-2 infection and causing high morbidity. We will use primary cells from different donors pertinent to different risk factors and correlate them with our experimentally determined parameters of infection and immune response. We will then validate our approach using circulating strains of SARS-CoV-2 isolated at the CHU Bordeaux (Hopital Pellegrin) from patients with known history.
